ABSTRACT: Paraoxonase-l (PON.1) is widely distributed among
tissues of different organs such as liver, kidney and intestine. It is present
in the serum associated with HDL-C. Overproduction of nitric oxide (NO)
has a contradictory effect on the inflammation. The present study aimed to
investigate the effect of garlic and N(j)-nitro-L-argininemethyles.ter (LNAME)
on PONl activity in acetic acid induced colitis. Male rats were
treated with garlic (orally) and L-NAME (I.P) at dose level 0.25 g/Kg and
10 mg/Kg respectively for 4 consecutive weeks and during induction oi
colitis.
Treatment of rats with acetic acid intra-rectally for three consecutive days
caused a significant macroscopic, microscopic degenerati(" ,1lcrease in
colon weight and decrease colon length. Pre-treatment of rats with garlic
or garlic plus L-NAME significantly reduced macroscopic and
microscopic colon degeneration.. They also significantly reduced the
increase in the colon weight and the decrease in the colon length induced
by acetic acid. Interestingly they significantly improved serum
paraoxonase activity and nitrate level, in addition to colonic tissue content
of MDA, GSH, SOD and CA T compared to colitis group. Conclusion.
the data indicated that garlic can protect rat colon against ulcerative colitis
induced by acetic acid. L-NAME has synergistic effect with garlic on
PONl activity and normalizes the serum nitrate level.
